小中大Should be like this:
1. In regular WB, usually you can not see IgG-H, or -L. Because the commercial conjugated-2nd ABs are optimized to avoid cross-reactions. But not absolutely, so be careful when you choose conjugated-2n-AB.
2. If IP, or Co-IP is applied. The thing becomes different. The L- or H-chain will appear, even MUST appear to prove your experiment works fine. Because: a) the 2n-AB recoginze the AB used in IP, or Co-IP; b) the 2n-AB re-nature with the AB used in IP, or Co-IP; and c) the 2n-AB cross-react with the AB used in IP, or Co-IP
