首先,不太赞同你的想法,准确的分析方法是保证制剂实验结果准确的前提,相关的方法学验证一定要做的,如果自己都对自己的分析结果心里没底,那相关的实验怎能保证有信心呢。而且有时文献上的数据也不一定准确的,影响胰岛素溶解度的因素有很多,其自身的结构或取代基团的差异就会导致溶解度的差异,所以自己检测一下还是有必要的,可以用少量体积的PBS溶解胰岛素,胰岛素少量多次加入,记录溶解的胰岛素的量,直到PBS不能再溶解胰岛素为止,这样可以大致的确定溶解度,离心后再进一步用液相检测浓度就行了。
这个溶解度数据很难直接在文献上查到的,一般文献上都是直接给出胰岛素释放度的数据,根本没有原始数据,对你有参考价值的文献能找到两篇:
