Abstract:
Absorption of 14C-labeled ambroxol (I-HCl) [23828-92-4] after oral administration was fast and complete. Elimination half-life of radioactivity in the blood was 20-25 h in rats, dogs, and man and only 2 h in rabbits. This apparent elimination half-life was governed by the disposition of acidic metabolites of I. In man and rabbits radioactivity was excreted almost completely into the urine, whereas in rats and dogs biliary excretion was also obsd. Routes of biotransformation were similar in all 4 species. I was metabolized first to NA 873 (II) [18683-95-9] and finally to dibromoanthranilic acid [609-85-8]. Conjugation of I and metabolites was obsd. mainly in man and rabbits.
