(收 稿 日 期 :20 04- 07 -04 ) 【研究报告】 〔文章编号〕1004-8685(2004)06-693-03 沉淀缓冲增菌法提高水中ElTo :霍乱弧菌阳性检出率的研究和应用‘ 陈德云,张慧红,胡伟宁,陈国雄,符发雄,巫结冰,老倩群 (佛山市顺德区疾病预防控制中心,广东528300) 摘 要 〔目的〕应用化学沉淀剂将ElT or霍乱弧菌沉淀聚集于水体下层,在缓冲溶液作用下保持特定的生长环境, 从而起到抑制其它肠杆菌的生长,避免和减少检测中易出现假阴性的误差,以达到提高霍乱弧菌的阳性检出率。为疫情 监测和调查提供准确可靠的科学依据。〔方法〕以碳酸钠为介质,硫酸亚铁为沉淀剂,在一定比例用量下,沉淀剂将水中 霍乱弧菌沉淀聚集于水体下层,弃去上层清液沉淀中加入碳酸盐缓冲溶液。用等量双倍浓碱蛋白陈进行增菌、分离、鉴 定。(结果〕沉淀剂可将水中97.67%一99.85% El Tor霍乱弧菌沉淀至水体下层,缓冲溶液在增菌过程中声变异数 1.19%,处于不变的良好状态。在此条件下El Tor霍乱弧菌最小检量0.014 - 0.140cfu/ml,比常规增菌法阳性检出提高一 个对数;沉淀缓冲增菌法比常规增菌法阳性检出率提高2.6倍。检测中无需二次增菌,比常规增菌法缩短一半的工作周 期,同时可降低节约70%的试剂成本。〔结论〕本检验方法具有操作简便,精确度高,最小检出量大,阳性检出率高,工作 周期短,试剂成本低的特点,适用于霍乱传染病的预防控制和调查。 关键词霍乱弧菌;沉淀缓冲;阳性检出率 Study and application of precipitation bufering proliferation method to enhance positive rate of El Tor vibrio cholerae in water ChenD eyun,ZhangH uihong,Hu W eining,et al .ShundeC enterfo rD iseaseC ontrolan d an was used to precipitate El Tor Vihrio Prevention, Guangdong 528300, China.
