Cytogenetic Analysis: Chromosome studies indicate a modal number of 63 with 3 distinct marker chromosomes and a small ring chromosome. This is a hypertriploid human cell line. The modal chromosome number was 61, occurring in 32% of cells.
However, cells with 63 chromosomes also occurred at a high frequency (22%). The rate of cells with higher ploidies was 8.5%.
Isoenzymes: G6PD, B

Comments: Growth is inhibited by 1 unit/ml L-asparaginase.
The cells will grow in soft agar.
Propagation: ATCC complete growth medium: Dulbecco's modified Eagle's medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate and 4.5 g/L glucose, 90%; fetal bovine serum, 10%

Receptors: insulin, expressed
glucocorticoid, expressed
Tumorigenic: Yes, in athymic mice
Strain: Wistar
Comments: Secretory activity is inducible by glucocorticoid stimulation, and is accompanied by extensive re-organization of the endoplasmic reticulum.
Propagation: ATCC complete growth medium: Ham's F12K medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 80%; fetal bovine serum, 20%
Growth Conditions: The cells cells grow slowly, in clusters. They tend to pile up and appear refractile.
Temperature: 37.0 C Atmosphere: air, 95%; carbon dioxide (CO2), 5%

Subculturing: Protocol: Monolayer never becomes confluent. Subculture when patches of cells start forming "domes".
Remove and discard culture medium.
Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.53 mM EDTA solution to remove all traces of serum that contains trypsin inhibitor.
Add 2.0 to 3.0 ml of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37癈 to facilitate dispersal.
Add 6.0 to 8.0 ml of complete growth medium and aspirate cells by gently pipetting.
To remove trypsin-EDTA solution, transfer cell suspension to centrifuge tube and spin at approximately 125 xg for 5 to 10 minutes. Discard supernatant and resuspend cells in fresh growth medium. Add appropriate aliquots of cell suspension to new culture vessels.
Incubate cultures at 37癈.

Cytogenetic Analysis: hyperdiploid; modal number = 73; range = 65 to 75; several translocations were evident; two to three apparently normal copies of number 7 (to which the cystic fibrosis gene has been mapped) were present in every metaphase examined
Age: 26 years
Gender: male
Ethnicity: Caucasian
Comments: This line was derived from a ductal adenocarcinoma (liver metastasis) from a patient with cystic fibrosis.
The cells exhibit ion transport activities consistent with cystic fibrosis and express the product of the CF gene (cystic fibrosis transmembrane regulator, CFTR).
CFPAC-1 cells show no effect of cAMP agonists, adenyl cyclase stimulators or phosphodiesterase inhibitors on Cl- flux, but do respond to Ca++ ionophores with increase Cl- efflux.
The cells have the most common form of the CF mutation, deletion of three nucleotides resulting in the absence of phenylalanine at position 508.
CFPAC-1 cells have epithelial morphology and polarization with apical microvilli, tight junctions and gap junctions.

Thank biotime for your help !
I have no time to wait for the cells from ATCC. I had the experince to buy a cell line from American, and waited about four months for dead cells. It 's a unpleasant experince.
Thanks anyway !
