大豆胰蛋白酶抑制剂sigma有type-1和type-2两种,请问他们是怎么分类的,和kunitz,bowman-birk有什么关系?他们对胰蛋白酶,凝乳蛋白酶和弹性蛋白酶抑制的特异性如何

根据蛋白酶抑制剂所作用于蛋白酶的类型,大致可以将其分为四大类:丝氨酸蛋白酶抑制剂、半胱氨酸蛋白酶抑制剂、天冬酰胺蛋白酶抑制剂和金属蛋白酶抑制剂。丝氨酸蛋白酶抑制剂是研究较多的一大类,其中就包括您提到的Kunitz和Bowman-birk这两个比较重要的抑制剂家族。

一般来说Kunitz型抑制剂分子量较大,属单头抑制剂(inhabition rate=1:1);而Bowman-birk型抑制剂分子量较小(8-12kD),在单子叶植物中多为单头抑制剂,在双子叶植物的多为双头抑制剂。大豆胰蛋白酶抑制剂的确可以分为Kunitz型和Bowman-birk型两类,其中后者可同时抑制弹性蛋白酶和胰蛋白酶。但不知道是否对应于您提到的sigma的“type-1”和“type-2”型。

This product is a 1% solution of trypsin in water.
Soybean trypsin inhibitor was first isolated by Kunitz.2
Several other related inhibitors are also found in
soybeans.3 Trypsin inhibitor from soybeans is a
monomeric protein containing 181 amino acid residues
in a single polypeptide chain crosslinked by two
disulfide bridges.4,5,6 The molecular weight
determined from the amino acid sequence is 20.1 kDa.
Soybean trypsin inhibitor inhibits trypsin, and to a
lesser extent chymotrypsin8 and plasmin.9 Soybean
trypsin inhibitor will also inhibit proteases with
mechanisms similar to trypsin. This product will also
inhibit plasma kallikrein and coagulation Factor XA.
However, this product will not inhibit metalloproteases,
tissue-based kallikrein, acid proteases, or thio
proteases.
Soybean trypsin inhibitor forms a 1:1 stoichiometric
complex with the protease active site. Upon formation
of this complex, trypsin may cleave a single arginineisoleucine
bond on the inhibitor.10,11 Inhibition is both
reversible and pH dependent. Dissociation of this
complex may yield a modified or native form of the
inhibitor.12 The optimal pH for trypsin binding is 8.0
with an association constant of greater than 109 at
pH 8.0 and 0.15 - 2.6 x 104 at pH 3.6 to 4.4.13

是sigma的产品中有Soybean trypsin inhibitor 后标有type 1,type2,所以我没法区分,不知道什么意思

从这两个产品的酶学登记号(EC number)及其他编号来看,这两个“type I-S”"type II-S”显然是同一个物质;根据产品注释,二者可能只是试剂纯度与用途不同,如前者1mg可抑制1-3mg胰蛋白酶,并且可用于细胞培养;而后者的抑制活力要低于前者。另外看到一个国内公司代理的这两种试剂,I型的价格也的确比II型高出许多倍,不知是否与此有关(cuturl('http://www.htc-bio.com.cn/pic/list16.asp?CurrPage=7'))。所以我想这两个类型只公司的产品编号,而不是对应于Kunitz或B-K两种不同的抑制剂类型。以上均为个人的理解,建议直接向代理公司或Sigma询问。

Thank you for contacting Sigma-Aldrich Technical Service. The type designations for these two product are Sigma in-house designations and do not represent any kind of biochemical significance. They are just used to differentiate between the two products. T9128 is a crude preparation of trypsin inhibitor (1 mg will inhibit 1 mg of trypsin with an activity of 10,000 BAEE units/mg protein). T6552 is of higher purity (1 mg will inhibit 1.0-3.0 mg ot trypsin with an activity of 10,000 BAEE units/mg protein), but more importantly, it has been suitability tested for use in cell culture applications (inhibition of trypsin after cells have been trypsinized).
