看完后应该就明白为什么qPCR产物要比较短,如果无法设计成比较短的产物,应该如何处理等等。(第一篇google一下篇名能查到,第二篇是免费的)
我觉得做qRT-PCR比常规跑胶传统RT-PCR还要省事,并且不光是省事,更重要的是准确。传统RT-PCR的一些要求在qRT-PCR上照样要求,并且省去了传统RT-PCR的好多要求,如:循环数的选择、上样量的准确、成像条件的一直等等可能干扰半定量准确性的因素,因为这些因素已经在qPCR中不存在了。
建议大家还是看一下qPCR的数学原理和化学原理,从理论上解决问题。
再者,对使用的qPCR仪性能和条件要有一定了解,比如,摄影家虽然对很多照相机都熟悉,但还是用自己熟悉的相机才能拍到好的相片,并且,相机只是工具,关键还是摄影家对摄影的理解。作者: loli 时间: 2015-8-3 18:19
原理如下:
The assay takes advantage of an environmentally sensitive fluorescence dye, such as Sypro Orange, and follows its signal changes while the protein undergoes thermal unfolding. When Sypro Orange is added to a properly folded protein solution, it is exposed in an aqueous environment and its fluorescence signal quenched. As the temperature rises, the protein undergoes thermal unfolding and exposes its hydrophobic core region. Sypro Orange then binds to the hydrophobic regions and becomes unquenched.
