小中大这样能达到良好的甲酯化效果么?
最近在做细菌脂肪酸的提取和测定,需要将脂肪酸甲酯化后跑GC-MS,查阅了相关的文献,发现用的最多的是Morrison and Smith(1964)的方法,即三氟化硼甲醇溶液甲酯化,文献中有这么一段话
Boron fluoride-methanol reagent was added under nitrogen, in the proportions 1 ml reagent per 4-16 mg of lipid, and the tube was closed with the screw cap. The tube was then heated in a boiling water bath for the requisite time, cooled, and opened. The esters were extracted by adding 2 volumes of pentane, then 1 volume of water, shaking briefly, and centrifuging until both layers were clear.
三氟化硼不是和水剧烈反应么,这里加水会不会和三氟化硼反应啊
还有就是这篇文献就是将三氟化硼甲醇溶液加到脂肪酸里拧上盖子100摄氏度水浴2min就可以,这和国标用冷凝管回流什么的简化了不少,能达到良好的甲酯化效果么,我看到有的文献还有在瓶子里拧上盖子100度水浴40min的 = =
