用流式细胞仪测定草的DNA含量

  这几天一直给别人做FCM,测定几种草中的DNA含量(pg/2C),一开始还是很明白的,可是随着实验前进,慢慢我也糊涂了,两次的实验结果完全相反,我都不知道哪里错了。重新配置了所有药品,结果都和第一次不一样,可是只有第一次里面有我要的数据啊?难道传说中的偶然?这次测定主要的方法来自”Determination of the level of variation in polyploidy among Kentucky Bluegrass Cultivars by Means of Flow Cytometry“.

  Introduce FCM–Flow cytometer

  Flow cytometry is an analytical technique that can be used to study many different aspects of cytology and genetics. For expample, it can be use to study the cell cycle by determining the proportions of cell in G1,S,and G2/M stages of the cell cycle, thus allowing the calulation of cell cycle times(Marie and Brown, 1993). It is also useful in performing task such as chromosome sorting and quantification of DNA content. Thus, flow cytometry enables a quick estimation of DNA amount by quantifying nuclear DNA content. Flow cytometry has the advantage fo being faster and less labor intensive than other methods of DNA content estimation, such as the microdensitometer technique and cytological chromosome counts.
